PROMETHEUS Clinical Trial Protocol: Tailoring Healthy Ageing with Lifestyle and Nutraceuticals
NUS Academy for Healthy Longevity, National University of Singapore
The published protocol states that DoNotAge.org provided SIRT6Activator used in the study. The study is funded by the XPRIZE Foundation and National University of Singapore.
PROMETHEUS is a multimodal healthy-ageing protocol that includes 2.4 g/day SIRT6Activator alongside exercise, dietary and sleep interventions, whey protein, creatine and other personalized components. Because the intervention is multimodal, outcomes cannot isolate SIRT6Activator efficacy.

DoNotAge Finished-Formulation Trial 2 — Whole Blood NAD+ Pilot
DoNotAge.org
DoNotAge.org recruited/enrolled participants, provided the finished formulation and placebo, and monitored adherence; whole-blood NAD+ analysis is described as performed by LifeLab1 as an independent laboratory service provider.
The hosted report describes a 28-day randomized, double-blind, placebo-controlled pilot in 24 enrolled healthy adults, with 19 completers, assessing whole-blood NAD+ levels.

Effects of Quercetin Supplementation on Cardiometabolic Parameters — NCT06230861
University of Central Lancashire
ClinicalTrials.gov lists DoNotAge.org as a collaborator; the University of Central Lancashire is the sponsor.
ClinicalTrials.gov describes an estimated 40-participant randomized, parallel, double-masked study testing quercetin against placebo with systolic blood pressure as a primary outcome and additional cardiometabolic, wellbeing and sleep measures. DoNotAge.org is listed as collaborator; no results are posted in the registry record.

2026Preclinical replication experimentOngoing preclinical replication. The first 2026 composition did not reproduce a significant grip-strength/rejuvenation effect; protocol ambiguities were later clarified and additional rat work was planned for November 2026, with separate mouse work also planned.
ICR–Unicamp Rat Rejuvenation Replication Experiment
Rejuvenation Science Institute (ICR) and University of Campinas (Unicamp)
ICR states that DoNotAge.org committed US$25,000 toward the rat rejuvenation experiment and describes DoNotAge.org as a project partner.
DoNotAge contributed US$25,000 to this ICR/Unicamp replication effort. The first old-rat test in 2026 did not show a significant grip-strength advantage versus controls. Later clarification of preparation, fraction-selection and dosing details prompted plans for further replication rather than a claim of successful reproduction.

DoNotAge SureSleep 21-Day Real-World Customer Study
BioStackr / commissioned by DoNotAge.org
DoNotAge.org commissioned the study in existing SureSleep customers. The company study page states that BioStackr designed the protocol, managed data collection, set exclusion rules and performed the analysis, with no commission or affiliate revenue from SureSleep sales.
The published study page describes 60 confirmed participants and 51 analysed completers across 13 countries, with 1,472 daily check-ins over 21 days. Outcomes were self-reported and compared each participant's final five days with their own baseline. The study had no placebo arm, randomisation or blinding.

DoNotAge-Funded PS18 Exercise-Mimetic Study in Aged Mice
USC Leonard Davis School of Gerontology / Cortes Lab
DoNotAge funded the aged-mouse experimental step presented by Constanza Cortes at DoNotAge Conference 2026. The project tests the prosaposin-derived peptide PS18 against sedentary control and treadmill exercise; it is not a human trial.
Conference materials describe an eight-week DoNotAge-funded aged-mouse experiment comparing sedentary vehicle control, sedentary PS18 and treadmill exercise, with frailty/healthspan, cognition, metabolic, body-composition and tissue outcomes. Published Cortes-lab skeletal-muscle TFEB/muscle-to-brain studies provide mechanistic rationale but are separate experiments and do not constitute PS18 efficacy results.

POLYamine Treatment in Elderly Patients With Coronary Artery Disease (POLYCAD) — NCT06186102
Aarhus University Hospital
The peer-reviewed POLYCAD protocol states that DoNotAge donated the spermidine intervention and placebo capsules and had no role in the study design, conduct, analysis or reporting.
POLYCAD randomizes 187 adults aged 65 years or older with coronary artery disease to 24 mg/day spermidine or placebo for 48 weeks. Co-primary outcomes include left-ventricular mass by cardiac MRI, VO2peak, appendicular lean mass and hs-CRP.
